摘 要:目的 探讨染色体7q35区醛糖还原酶(AR)基因启动子区C- 10 6 T多态性与2型糖尿病肾病(DN)相关性。方法 运用聚合酶链反应限制性片段长度多态性技术(PCR- RFL P)、DNA测序技术及高浓度琼脂糖凝胶电泳分离技术对AR基因启动子区C- 10 6 T多态位点的等位基因和基因型进行了检测,将2型DM患者139例,按尿微量白蛋白排泄率(UAER)分为早期DN组(DN+组,6 1例)和非DN组(DN-组,78例)两个亚组,并以6 3例健康成人作为对照(CON组)。结果 AR基因启动子区C- 10 6 T多态位点存在T、C两种等位基因和CC、CT、TT三种基因型,DM组和CON组间各等位基因和基因型频率差异无显著性,DN+组T等位基因和CT基因型频率明显高于DN-组和CON组(P<0 .0 5 )。结论 AR基因启动子区C- 10 6 T多态性与DN的发生、发展有关。Objective To investigate the associatio n between polymorphism of the C-106T at promotor region of aldose reductase(AR) gene in chromosome 7q35 and nephropathy with type 2 diabetes in Northern Chines e.Methods A case-control s tudy for 202 Chinese (including 139 type 2 diabetes mellitus with or without ne phropathy and 63 normal control subjects) was performed.The genotypes and all eles of polymorphism of C-106T at promotor region of AR gene were determined by polymerase chain reaction restriction fragment length polymorphism(PCR-RFLP) met hod and combined with 4% agarose gel electrophoresis.Results The frequency of the T allele and CT genotype at promotor region of AR gene in DN+ group were sig nificantly higher than that in DN- and control(CON) group(P <0.05),but th ere was no significant difference between DM and CON group (P >0.05).Conclus ion The T alleles or CT genotype at promotor region of AR gene are related to nephropathy with type 2 diabetes.
