人体血液及脑脊液中头孢吡肟浓度的HPLC测定方法研究

摘　　要：目的:建立高效液相色谱法测定血液和脑脊液中头孢吡肟的药物浓度。方法:待测样品经蛋白沉淀处理后,内标法进行高效液相紫外检测,选用三氯乙酸作为蛋白沉淀剂,头孢他啶作为内标;色谱柱采用Intertsil ODS-3(5 μm,4.6 mm×250 mm)色谱柱;流动相为:10%乙腈(v/v),20 mmol·L^(-1)醋酸铵,用冰醋酸调pH至4.00:检测波长为254 nm;结果:在选定的色谱条件下,头孢吡肟与血液及脑脊液中的杂质分离良好,头孢吡肟的保留时间在6 min左右。血液中和脑脊液中头孢吡肟的线性范围分别为0.5-200μg·mL^(-1)和0.1—100μg·mL^(-1),回收率分别为76.17%-97.55%和82.28%-92.10%,最低检测限分别为0.5μg·mL^(-1)和0.1μg·mL^(-1)。结论:该法可以用于测定血液及脑脊液中的浓度,操作简便,灵敏度高,结果准确,重现性好。Objective:To establish an HPLC method for determining concentration of cefepime in human serum and cerebrospinal fluid(CSF).Method:The sample was deproteinated by trichloroactic acid and determined with HPLC and ultraviolet detection.The internal standard was ceftazidime.The analytical column was Intertsil ODS -3(5μm, 4.6 mm×250mm).The mobile phase consisted of 10% acetonitrile and 20mmol·L^(-1) ammonium acetate(pH= 4.00).Detection was performed at 254nm.Results:Under the selected chromatographic conditions, cefepime was depart from the foreign matter of blood and CSF.The observed retention time for cefepime was about 6 min.The linear range of cefepime was 0.5-200μg·mL^(-1) in plasma and 0.1-100μg·mL^(-1) in CSF.The recoveries of cefepime in plasma and CSF were 76.17% -97.55% and 82.28% -92.10% respectively.And the detection limit were 0.5 μg·mL^(-1) and 0.1μg·mL^(-1) Conclusion:The assay method was shown to be suitable for determining the concentration of cefepime in plasma and CSF.This method is simple, accurate, sensitive and reproducible.
