摘 要:目的:探讨在维生素D3的作用下,白血病细胞株K562与SHI-1ppGalNAcTs表达的变化。方法:应用RT-PCR法研究在不同浓度维生素D3的作用下,白血病细胞株K562与SHI-1ppGalNAcTs表达的变化。结果1,25-(OH)2D3(10-6-10-8mol/L)可分别上调K562细胞株ppGalNAcT2、T4、T5的表达。但在SHI-I细胞株,随着1,25-(OH)2D3浓度的增加ppGalNAcTI表达增加,T3表达没有变化、T4表达减少。结论:白血病细胞株K562与SHI-1PP-GMNAcTs的表达不同。在不同浓度维生素D3的作用下,白血病细胞株K562与SHI-IppGalNAcTs表达的变化也不同,说明不同ppGalNAcTs对1,25-(OH)2D3的反应不同。Objectives: To observe the expression of polypeptide: N-acetylgalactosaminyltransferaseTs (ppGalNAcTs) in different leukaemia cells including K562 cell line and SHI-1 cell line and to study the variation of ppGalNAcTs expression after 1,25( OH)2D3 added to the culture medium. Methods: Use RT-PCR to detect(1) the mRNA expression of ppGalNAcTs in K562 cell line and SHI-1 cell line; (2) the expression discrepancy of ppGalNAcTs in K562 cell line and SHI-1 cell line after 1,25 ( OH)2D3 added to the culture medium. Results: 1. The expression of ppGalNAc was different in two leukaemia cell lines. 2. After 1,25(OH)2D3 added to the culture medium,the mRNA expressions of ppGalNAcT2,T4,T5 in K562 cell line increased,and ppGalNAcTl in SHI-1 cell line increased too. But ppGalNAcT3 have no change,and pp-GalNAcT4 in SHI-1 cell line decreased. Conclusion: This study showed that the expression of ppGalNAcTs in different leukaemia cell was different. This result suggested that different ppGalNAcTs had different particular substances. The mRNA expression of ppGalNAcTs was defferent after 1,25(OH)2D3.
