系统性红斑狼疮患者外周血Vα24-Vβ11自然杀伤T细胞致病性的研究

摘　　要：目的研究系统性红斑狼疮(SLE)患者外周血Vα24Vβ11自然杀伤T细胞(NKT)细胞数量变化和功能状态在致病中的作用。方法采用流式细胞仪检测32例SLE患者和30名正常对照组Vα24Vβ11NKT细胞数量以及体外活化前后表达血细胞分化抗原(CD)69和白细胞介素4(IL4)、Ⅱ型干扰素(IFNγ)的水平。结果SLE患者外周血Vα24Vβ11NKT细胞数量(044%±025%)低于正常对照组(107%±023%,P<001),NKT细胞体外活化前CD69表达为526%±212%,正常对照组为1147%±286%(P<005);活化后表达CD69为5661%±047%,正常对照组为9671%±033%(P<001)。SLE患者NKT细胞活化前其胞浆内IL4含量为3246±549pg/ml,正常对照组为1221±331pg/ml,活化后其胞浆内IL4含量为4838±853pg/ml,正常对照为2693±684pg/ml(均P<005),而IFNγ含量活化后为1932±645pg/ml,正常对照组为3365±1191pg/ml(P<005)。结论SLE发病可能与NKT细胞数量的低下及功能严重失调相关。Objective To explore the changes of amount and function of Vα24-Vβ11 natural killer (NKT) cells in peripheral blood of patients with systemic lupus erythematous (SLE). Methods The amount of Vα24-Vβ11NK T cells and expression rate of cluster of differentiation (CD) 69, interleukin4 IL-4 and interferonr (IFN)-γon these cells in 32 cases of SLE and 30 healthy persons were tested by flow cytometry.Results The amount of Vα24-Vβ11NK T cells in SLE group was 0.44%±0.25% and that in control group was 1.07%±0.23%. The expression rate of CD69 in SLE group was 5.26%±2.12% and that in control group was 11.47%±2.86% before cell activation. It was 56.61%±0.47% and 96.71%±~0.33% respectively after cell activation. INF-γon NK T cells in SLE group was 19.32%±6.45 pg/ml and lower than that in controls (33.65±11.91 pg/ml,P<0.01 or P<0.05). Whereas the expression rate of IL-4 were 32.46 ± 5.49 pg/ml before cell activation and 48.38 ± 8.53 pg/ml after cell activation, which were higher than those in control group(12.21 ± 3.31 and 26.93 ± 6.84 pg/ml respectively,P<0.01).Conclusion Pathogenesis of SLE may be related to the amount of NK T cells and their serious maladjustment.
