摘  要:目的对比有饲养层的有血清培养体系(feederlayer,serum—containingmedium ,FSCM)和无血清培养体系(serumfreemedium ,SFM)培养人角朊细胞(keratinocyte ,KC)的特征。方法以两种不同培养体系培养人KC ,比较2 4h细胞贴壁数、完全汇合时间和最终分化形式。结果KC在两种不同培养体系中的形态、2 4h细胞贴壁数和完全汇合时间无统计学差异。FSCM体系能促使角朊细胞发生终末分化;SFM体系内角朊细胞增殖较FSCM体系快,但未发生终末分化。结论有饲养层的有血清培养体系培养人角朊细胞形成复层膜片,适合临床应用;无血清培养体系培养人角朊细胞形成单层细胞,适合KC生物学性征的实验研究。Objective To demonstrate the characterization of keratinocytes cultured in feeder cell layer, serum-containing medium(FSCM) and in a serum-free medium(SFM). Methods The adhering numbers of KC, the time of confluence and the differentiation of keratinocytes in two different culture systems were compared. Results The morphologiy pictures of cells were similar ,and there was no statistical difference in adhering numbers and the time of confluence in the two different culture conditions, the KC in FSCM system differentiated into multilayer stratified epithelium, while the KC in SFM system quickly proliferated but stratification was absent. Conclusion KC cultured under FSCM can be used for grafting, while that cultured in a serum-free medium may be applied for the study on the biological properties.
