摘 要:目的 研究胆道梗阻及再通大鼠血清GH的变化规律及在肝细胞胆红素代谢中的作用。方法 对胆道梗阻及再通大鼠动物模型的72份血清样本进行放免检测GH;肝脏组织样本行原位杂交染色在肝细胞胞浆染色定位 STAT5,计算机图像分析测定,半定量比较肝细胞胞浆STAT5表达情况,并与血清GH、血清胆红素作相关分析。结果 在胆道梗阻时机体存在GH分泌、释放不足从正常时的(6.54±1.09)下降到14d的(1.97±0.64),再通后GH缓慢恢复;STAT5在胆道梗阻大鼠肝细胞胞浆中表达(阳性面积比)明显下降由正常(0.335±0.09)到14d(0.045±0.02),再通后恢复至正常水平,图像分析、半定量比较发现其随血清GH、胆红素的升高或降低而发生变化。结论 胆道梗阻时机体存在GH分泌、释放不足,并通过STAT5信号途径影响肝细胞代谢胆红素。Objective To study the changes of serum GH and the role in bilirubin metabolism of liver in bile duct obstruction-restoration of bile flow rats.Methods Wistar rats of 14d after common bile duct ligation(CBDL) were subjected to restoration of bile flow(RBF) for tests and sham operations (SHAM)for control. The samples were collected on 3,7,14d after RBF.Serum GH in bile duct obstruction-restoration of bile flow rats was measured by radioimmunoassay, STAT5 activation of liver tissue by ISHH, correlated with GH and bilirubin in serum.Results Serum GH was markedly lower in obstructive jaundice rats.After restoration of bile flow,GH increased gradually , related with the bile flow restoration time. The changes of serum GH positively correlated with STAT5mRNA of liver tissue and negatively with bilirubin in serum.Conclusion Serum GH was markedly lower in obstructive jaundice rats.Bilirubin metabolism of liver was regulated by GH/STAT5 signaling pathway.
