摘 要:目的 :研究急性吗啡中毒大鼠主要脏器内吗啡的分布 ,建立生物检材吗啡的固相萃取 (SPE)和高效液相色谱法 (HPLC)测定方法。方法 :以Φ4.6×150mmC18色谱分析柱 ,甲醇 -10mmol·L -1庚烷磺酸钠 (50:50)为流动相 ,检测波长233nm。结果 :平均回收率为98.53%,RSD2.4% ;最低检测浓度为5ng·ml-1。1例吗啡中毒死亡者的体内分布为血1.74μg·ml-1、脑0.14μg·g-1、肝0.21μg·g-1、肾0.08μg·g-1,肺0.02μg·g-1,尿液5.84μg·ml-1。结论 :此法工作曲线稳定 ,灵敏度高 ,操作步骤简单 ,并应用于吗啡类毒品中毒的法医毒物分析。血、尿是中毒者毒物分析得较好检材。Objective:To study the distribution of morphine in rat's oranges,during acute poison and.to determination the morphine in biological tissue specimen by high performance liquid chromatography(HPLC).Methods:TheΦ4.6×150mm C18column was used.The moˉbile phase consisted of methanol-10mmol·L -1 sodium heptanesulfonate(50:50).Detection was monitored by ultraviolet absorption at233nm.Results:The average recovery of morphine was98.53%.RSD was2.4%.The lowest detectable concentration of morphine was5ng·ml -1 .The concentrations of morphine in man died due to morphine poisoning were as follows:blood1.74μg·ml -1 ,brain0.14μg·g -1 ,liver0.21μg·g -1 ,kidney0.08μg·g -1 ,lung0.02μg·g -1 ,urine5.84μg·ml -1 .Conclusion:The method is stable,high sensitive and easy operaˉtion.It has been used in forensic science application on analysis of morphine poisoned.Blood and urine are better samples for analysis of morphine poisoning victim.
