非甾体抗炎新药舒林酸的抗炎、镇痛和解热作用

摘　　要：舒林酸有明显的抗炎、镇痛、解热作用。灌胃给药使巴豆油致小鼠耳廓肿胀、角叉菜胶致大鼠足跖肿胀抑制２５％的半数有效量（ＥＤ５０）分别为３６±５６和１４±５ｍｇ／ｋｇ，２０和４０ｍｇ／（ｋｇ·ｄ）或连续５ｄ对棉球肉芽肿和佐剂性关节炎也有显著抑制作用。镇痛试验：小鼠热板法和醋酸扭体法测得ＥＤ５０分别为４６±１９和５２±１１ｍｇ／ｋｇ；使酵母致热大鼠降低１℃的ＥＤ５０为６．２±２．５ｍｇ／ｋｇ；使大鼠试验群５０％致胃出血的半数致溃疡量（ＵＤ５０）为５６±１２ｍｇ／ｋｇ。Anti-inflammatory,analgesic and antipyretic activities of sulindac were observed in laboratory animals.The anti-inflammatory activities of sulindac were evidenced bv inhibiting various acute and chronic inflammation models including croton oil topical inflammation of ear in mice,carrageenan induced paw edema,cotton pellet granuloma and adjuvant arthritis in rats.The ED50 of sulindac ig were determined in the following assays:croton oil topical inflammation of ear in mice(25% inhibition rate),36±s 6 mg/kg;carrageenan foot edema in rats,14±5 mg/kg.In anti-analgesic studies using hot plat test and writhing test in mice,the ED50 werc 46±19 and 52±11 mg/kg respectively.In the antipyretic test to lower body temperature of rats by 1℃yeast-induced fever the EDs0 was 6.2±2.5 mg/kg.The UD50 of sulindac required to cause haemorrhage of the gastric mucosa in 50% of rats was 56±12 mg/kg.
