摘  要:目的:探讨急性心肌梗死(AMI)后大鼠脾淋巴细胞对体外培养心肌细胞的杀伤作用。方法:SD大鼠90 只,随机均分为AMI组,假手术组和正常对照组。在体结扎SD大鼠冠状动脉左前降支(LAD),建立AMI大鼠模型 和假手术对照。手术后第1周,2周和4周分别分离大鼠脾脏淋巴细胞,与乳鼠心肌细胞共培养,结晶紫法测定淋 巴细胞对心肌细胞的杀伤效应。结果:AMI后2周和4周大鼠淋巴细胞对培养心肌细胞的杀伤效应((36.4± 7.2)%,(68.2±7.1)%)较假手术组((22.5±4.5)%,(34.7±6.3)%)和正常大鼠组((16.7±3.2)%和(17.6± 2.7)%)明显增强,1周时3组杀伤效应差异无统计学意义。结论:AMI后大鼠的淋巴细胞能够杀伤正常的心肌细 胞,该机制可能参与了AMI后免疫机制介导的心肌损伤和心室重塑过程。Aim: To explore the changes of rat lymphocyte cytotoxicity against cultured cardiac myocytes in vitro after acute myocardial infarction(AMI). Methods: A total of 90 SD rats were allocated into AMI group,sham operation group(SH),and normal control group,30 rats in each group.AMI model was adopted by in vivo ligation of left anterior descending branch in SD rats. One, two and four weeks after operation, lymphocytes were obtained from rat spleens and incubated with neonatal rat cardiac myocytes. Crystal-violet staining assay was used to assess lymphocyte cytotoxic activity against cultured cardiac myocytes. Results: Two weeks and four weeks after operation, lymphocyte cytotoxic activity were significantly higher in AMI group((36.4±7.2)% and (68.2±7.1)%)) compared with SH-MI ( (22.5±4.5)% and(34.7± 6.3)%) and normal control group (16.7±3.2)% and(17.6±2.7)%). Conclusion: Lymphocytes from AMI rats can recognize and kill normal cardiac myocytes in vitro, which perhaps participate in the autoimmunization mediated cardiac myocytes injury and ventricular remodeling processes after myocardial infarction.
