摘 要:目的:探讨消渴灵浓缩液对糖尿病大鼠坐骨神经IGF 1mRNA表达的影响。方法:将30只链脲佐菌 素诱导的糖尿病大鼠模型随机分为模型组、西药对照组、中药治疗组,每组10只。另选10只鼠龄、体重相匹配的 大鼠作为正常组。采用相对定量逆转录聚合酶链式反应(RT PCR)检测病程8周后各组大鼠坐骨神经IGF 1mR NA表达的水平。结果:中药组、西药组、正常大鼠组坐骨神经IGF 1mRNA相对表达量均增加,差异无统计学意义 (P=0.213,P=0.822,P=0.304);模型组坐骨神经IGF 1mRNA相对表达量下降,与中药组、西药组、正常大鼠 组比较,差异均有统计学意义(P<0.05)。IGF 1mRNA表达水平与血糖呈负相关(P<0.05)。结论:糖尿病大鼠 坐骨神经组织IGF 1mRNA的表达下降,消渴灵浓缩液可上调糖尿病大鼠坐骨神经IGF 1mRNA的表达水平,可能 是消渴灵浓缩液防治糖尿病周围神经病变的作用机制之一。Objective To investigate the effect of xiaokeling concentration fluid on insulin-like growth factor-1(IGF-1) mRNA expression in sciatic nerve of Streptozotocin-induced diabetic rats. Methods Thirty diabetic rats were randomly divided into model group, mecobalamin tablets group, and xiaokeling concentration fluid group. The IGF-1 mRNA level in sciatic nerve of each group was determined after 8 weeks by relative quantity RT-PCR. Results The IGF-1 mRNA level in sciatic nerve of diabetic rats between xiaokeling concentration fluid group, mecobalamin tablets group and normal group showed no significant difference (P=0.213,P=0.822,P=0.304), while was significantly higher than that of the model group (P<0.05). IGF-1 mRNA level was negatively correlated with the level of blood sugar (P<0.05). Conclusion IGF-1 mRNA level decreased in sciatic nerve of diabetic rats. Xiaokeling concentration fluid can increase the IGF-1 mRNA level in sciatic nerve of diabetic rats. Xiaokeling concentration fluid is involved in the regulation of IGF-1 expression, and probably prevents diabetic peripheral neuropathy from deteriorating.
