摘  要:目的通过促分化因子的调节作用对神经干细胞向胆碱能神经元的定向分化进行探讨研究。方法采用细胞培养技术、免疫细胞化学方法观察新生大鼠皮层神经干细胞向胆碱能神经元定向分化的时空变化。结果新生大鼠皮层神经干细胞去除丝裂原后在促分化因子的作用下开始进行分化,至第7天可分化为成熟的胆碱能神经元,AchE反应明显阳性。在胆碱能神经元逐渐成熟的分化过程中AchE在细胞内的的阳性反应部位发生了变化,由开始时的核膜极其周围分布变化为成熟时的细胞质内分布。结论皮层神经干细胞在促分化因子的作用下可定向分化为胆碱能神经元,且AchE阳性反应具有时空变化特性。To study nerve stem cell directional differentiation to cholinergic neuron by adjusting action of promote differentiation factors. Observation the space-time change of cortex nerve stem cell in newborn rat orientation differentiation to cholinergic neuron by cell cultured and immunocyte chemistry methods. Cortex nerve stem cell of newborn rat began to differentiation after removed mitogen, and became cholinergic neuron in the seventh day,the response of Ach E was positive. The positive response position of Ach E in cell had changed during mature and differentiation stage of cholinergic neuron, and changed from distribution of initial nucleus to mature cytoplasm. [Conclusions] Cortex Nerve stem cell can directional differentiation to cholinergic neuron by promoting differentiation factors, and the Ach E positive response had characteristic of space-time change.
