摘  要:目的 通过研究重组人白介素11(rhIL-11)对正常IEC-6细胞增殖、细胞周期及其IL -1受体表达的影响,探讨IL -1对 小肠上皮细胞的调控作用以及相关机制。方法 利用MTT法检测rhIL -1对正常IEC-6细胞增殖的影响,通过流式细胞仪检测细 胞周期的变化,并采用Westernblot方法检测IEC-6细胞表面IL-11受体α链的表达情况。结果 rhIL-11可引起正常IEC-6细胞发 生增殖抑制和明显的G0/G1期阻滞,并诱导细胞表面IL 11受体α链表达增加。结论 rhIL-11可能通过与IEC-6细胞表面特异性受 体结合,从而启动胞浆内信号转导通路,引起细胞效应。Objective In order to investigate the effects of interleukin-11 (IL-11) on the small intestinal epithelial cells, we observed the effect of recombinant human interleukin-11 on the proliferation, cell cycle and the expression of IL-11 receptor of IEC-6 cells in vitro. Methods The cell cycle, proliferation and the expression of IL-11 receptor of IEC-6 cells were determined by means of MTT, flow cytometry and Western blot. Results The results demonstrated that rhIL-11 could induce proliferative inhibition and cell cycle retardation in normal IEC-6 cells. Moreover, rhIL-11 also evoked the expression of alpha-chain protein of IL-11 receptor in IEC-6 cells. Conclusion These results implicated that rhIL-11 might affect the IEC-6 cells by combining with the receptor on the cell membrane to regulate the signal transduction pathway in the cytoplasm.
