摘  要:目的 通过研究重组人白介素11(rhIL -1)对照射小鼠小肠上皮细胞凋亡及相关基因表达的影响,探讨IL-11在急性放 射病小鼠小肠上皮细胞辐射损伤中的治疗作用以及相关机制。方法 利用琼脂糖凝胶电泳法和原位末端标记法检测rhIL -1对 8.0Gy60Coγ射线照射小鼠小肠上皮细胞凋亡的影响,通过免疫组织化学的方法观察Bcl -和Bax的表达变化。结果 rhIL -1治疗 和预防给药可明显地抑制小鼠小肠上皮细胞凋亡的发生,并诱导Bcl 2表达而使Bax表达明显减弱。结论 rhIL-11可能通过影响 Bcl -和Bax的表达而抑制辐射引起的小肠上皮细胞凋亡,从而产生防护作用。Objective To investigate the effect of IL-11 on small intestinal epithelial cells and its underlying mechanism, the effect of recombinant human interleukin-11 on induction of apoptosis of epithelial cells of the small intestine and changes in its related gene expression were observed in irradiated mice. Methods Cell apoptosis of epithelial cells of the small intestine was identified with in situ terminal labeling and DNA electrophoresis. The expression of Bax and Bcl-2 in small intestine epithelial cells was assessed by immunohisto-chemistry. Results The results demonstrated rhIL-11 treatment and pretreatment could significantly prevent apoptosis of the crypt cells after irradiation, and the effects might be attributed to induced expression of Bcl-2 and depression of expression of Bax in the cells. Conclusion rhIL-11 could protect small intestine from radiation injury by preventing apoptosis of the crypt cells, which might be attributed to changes in the expression of Bcl-2 and Bax in the cells.
