Expression of Integrin α_v and Le^y Oligosaccharide in Endometrium of Peri-implantational Period in Mice under Different Conditions

摘  要:目的:探讨促排卵药、胚胎本身及孕后激素等条件对小鼠子宫内膜整合素av和Ley寡糖的表达及对着床窗开放的影响。方法:将成年雌鼠分为对照和实验两大组。对照组为自然受孕;实验组为用促排卵药后受孕,其中一部分为假孕,另一部分为事先行一侧输卵管结扎。各组分别于孕2-7 d取子宫标本2份,一份石蜡包埋行HE染色和免疫组化染色测定Ley寡糖,另一份于-80℃保存,冰冻切片行免疫组化染色测定整合素av。结果:①对照组于孕7 d,大部分小鼠子宫外观即见明显胚胎着床,显微镜下从孕5 d起出现子宫内膜间质的蜕膜反应,但实验组无1例有此变化。②整合素av在小鼠子宫内膜腺上皮细胞胞浆内表达。对照组的表达于孕4 d达高峰,而各实验组的表达高峰均推迟1-2 d,且表达水平较对照组明显减弱(P <0.05)。结扎侧和未结扎侧子宫内膜整合素av的表达高峰无明显差异。③Ley寡糖在小鼠着床期表达于子宫内膜腔上皮和腺上皮表面,其表达两组间无明显差异,不同妊娠天数之间的表达亦无明显差异。在对照组,于孕4 d开始Ley寡糖在子宫内膜间质有表达,孕6 d达高峰,而实验组未见一定的规律。结论:①小鼠子宫内膜腺上皮细胞胞浆内整合素av的表达与子宫内膜着床窗的开放一致。②促排卵药能抑制小鼠子宫内膜着床窗期整合素av的表达,延迟着床窗的开放;Objective: To investigate whether ovulation drugs, embryos and pregnant hormones can influ-ence the expression of integrin av and Ley oligosaccharide and/or the opening of implantation at window in en-dometrium of mice. Methods: The female mice were divided into two groups, control and experiment group. Thecontrol group consisted of pregnant mice ; the experiment group consisted of mice pregnant after inducing ovulation,normal part of which was pseudopregnant, and the others was pregnant after unilateral tubal ligation. The twouterine specimens of these groups were taken respectively during day 2-7, one went through HE stain and immuno-histochemistry stain to determine Ley oligosaccharide, the other was preserved under -80℃ for measurement ofintegrin av. The most cases of uteri on day 7 of pregnancy in control group showed obvious signs ofimplantation.Observation under microscope showed the decidual change of endometrial stromal cells on day 5 of pregnancy incontrol group, while there was no such changes in experiment group. 2) Integrin av was expressed in plasma ofendometrial gland epithelial cells. The peak of expression in control group was on day 4 of pregnancy, but the thatin experiment group was postponed by 1-2 days and the expression level decreased significantly (P<0.05). Therewas no difference in expression between the non-ligated and ligated uterine endometria. 3) Ley oligosaccharide wasexpressed on the surface of luminal and gland epithelial cells of endometria in implantational stage of mice. Therewas no difference in Ley oligosaccharide expression between the control and experiment group. There was nodifference during different pregnant ages, either. Interestingly in control group, Ley oligosaccharide began to ex-press in the endometrial stromal cells on day 4 of pregnancy, and thepeak of expression was on day 6 of pregnancy.But in experiment group, we cant find any rules of expression.
