鸡病原性大肠杆菌（血清型O_（78））纤毛亚单位苗免疫原性的研究

摘　　要：本实验利用透射电镜观察了鸡源大肠杆菌（血清型Ｏ＿（７８））表达的纤毛。提取并初步纯化了纤毛。通过ＳＤＳ－聚丙烯酰胺凝胶电泳检测了提取物的纯度及其亚单位分子量。表明提取物属Ｉ型纤毛。用提取纤毛制备油佐剂苗。评估了该苗的免疫原性，免疫组和阳性对照组病变级数有极显著差异（Ｐ＜０．０１），苗的保护指数分别为一次免疫６４．２８％，两次免疫７４．６０％。用ＥＬＩＳＡ检测了免疫后６８天鸡的抗体消长。通过统计分析二免后抗体水平和一免组同期抗体水平有显著差异。The presence of pili on E.Coli pathogenic to chicken was examined by transmissionelectron microscope (TEMS).The Purity and the molecular weight of the isolated piliwas verified by TEMS and sodium dodecyl sulfate polyarylamide gel electrophoresis(SDS-PAGE. The results indicate that the obtained pili was type I. An oilemlsified vaccine wasprepared with the isolated Pili. The inimunogenicity of the vaccine was envaluated. Lesionscored values of vaccinated group were significantly lower (P<0.01) than those of posi-tive controls.The protection index were 64.28% and 74.60% when the chickens iznmu-nised once and twice respectively.The antibody levels of chickens were measured withELISA 68 days post vaccination.The antibody level to pili of twice vaccinated cbickenswas significantly higher than that of once vaccinated ones.
