摘 要:目的:比较MTT分析法、空斑减数试验及细胞病变效应(CPE)观察3种方法在评估药物体外抗病毒效果分析中的优劣,为选择适宜的药物体外抗病毒效果的检测方法提供实验依据。方法:分别采用 CPE法、MTT分析法和空斑减数试验法评价大黄素于体外抗疱疹病毒 1 型、疱疹病毒 2 型和柯萨奇病毒增殖的效果,比较 3 种检测方法的结果,并分析该方法的实用性与利弊。结果:在抗疱疹病毒增殖的检测中,MTT法与空斑减数法所测结果具有显著性差异,空斑减数法比MTT法更准确,但对抗柯萨奇病毒增殖的检测,MTT法与空斑减数法无明显差异。结论:MTT法与 CPE法结合,可广泛用于对抗病毒药物的筛选,但对于疱疹病毒类非杀细胞病毒,使用空斑减数法较好。Objective: To compare the effect of plaque reductive assay, MTT assay and cytopathic effect (CPE) on evaluating the results of antiviral proliferation of medicines in vitro. Methods: Antivirus effects of Emodin on HSV-1, HSV-2 and Coxsackievirus B were assayed by plaque reduction assay, MTT assay and CPE, and the data were statistically analyzed by SPSS. Results: To evaluate the antiviral proliferation of Emodin, significance difference was found between the results of MTT assay and the plaque reductive assay for evaluating anti-herpes simplex virus. And the latter was more accurate than the MTT assay. But there was no difference between the plaque reductive assay and MTT assay in the research of anti-coxsackievirus. Conclusion: The results demonstrate that the MTT assay could be widely used in evaluation of antiviral activity, but plaque reduction assay can be more accurately to reflect the effect of medicine.
