摘  要:目的观察国际流行株HIV1env与IFNα2b基因共表达的颗粒化抗原疫苗与质粒DNA核酸疫苗免疫程序优化的效果。方法将重组质粒pJ16envIFNα2b经脂质体介导转染RK细胞,并在痘苗病毒中共表达HIV1env与IFNα2b基因,以间接免疫荧光与斑点酶联免疫法鉴定其表达产物。以表达产物与质粒DNA联合免疫鼠脾淋巴细胞,检测淋巴细胞转化率、细胞毒性T细胞杀伤活性、CD4+、CD8+细胞计数与体液免疫水平。结果pJ16envIFNα2b基因表达的颗粒化抗原疫苗具有良好的免疫原性,3vJ16envIFNα2b+pJ16envIFNα2b联合免疫效果优于vJ16envIFNα2b单独免疫。结论pJ16envIFNα2b基因表达的颗粒化抗原疫苗能诱导机体产生细胞免疫与体液免疫。Objective:To observe effect of HIV-1env and IFNα-2b genes coexpression vaccina and plasmid DNA vaccina Immunization Schedule.Methods:The plasmid pJ16env/IFNα-2b genes was transfected to RK cell under the mediation of liposome,and the expressed product was identified by indirect immunmofluorescence method and Dot-ELISA,The expressed product was used for immunizing mice combined with DNA vaccine,and the effect was evauated by lymphocyte transformation test,CTL,CD4+ and CD8+T cell counting as well as cellular and humoral response levels.Results:pJ16env/IFNα-2b vaccina showed good immunogenicity.The optimal Schedule was doses of pJ16env/IFNα-2b vaccina plus one dose of DNA vaccine.Conclusion:pJ16env/IFNα-2b genes coexpression proteins and both cellular and humoral immune responses were induced.
