摘　　要：目的制备TRAIL及其5种受体的特异性基因探针。方法以PHA刺激人外周血淋巴细胞(PBL)增殖,从而诱导TRAIL及其受体的mRNA转录水平提高,然后用RTPCR方法扩增出TRAIL及其5种受体的DNA基因片段,并采用基因测序法确证所得基因片段序列的正确性。结果获得了TRAIL及其5种受体的DNA基因片段。结论所用方法正确,为研究TRAIL及其受体调控细胞凋亡的机制提供了有价值的工具。Objective:To prepare the probes of human TRAIL and its five receptors.Methods:Human peripheral lymphocytes (PBL) were stimulated to proliferate by PHA. It can induce the gene expression of TRAIL and its receptors at the same time. All coding gene fragments of TRAIL and its five receptors were largely expanded by RT-PCR and isolated by electrophoresis and confirmed by gene sequencing.Results:The DNA fragments of TRAIL and its five receptors were obtained.Conclusion:The adopted method is right. It provides the useful tools for research in the regulating mechanism of the cell apoptosis induced by TRAIL and its receptors. [
