银屑病患者皮损中有丝分裂原激活的蛋白激酶的活性研究

摘　　要：目的　探讨有丝分裂原激活的蛋白激酶 (Mitogen -activatedproteinkinase,简称MAPK)在银屑病发病中的意义。方法　采用蛋白激酶磷酸化实验的方法研究了 1 3例寻常型银屑病患者皮损区、非皮损区和 1 3例正常人表皮中MAPK的激酶活性。结果　银屑病患者皮损区 (PL)中MAPK的活性为 3 0 . 46± 7 .2 1pmol/mgprotein/min ,银屑病患者非皮损区 (PN)为 1 6. 83± 3 .41pmol/mgprotein/min ,正常人表皮 (N)为 1 5 .3 1± 3 . 0 2pmol/mgprotein/min ,与PN和N相比 ,PL中MAPK的活性明显增高 ,而PN和N中MAPK的活性无明显差异。结论　MAPK可能参与了银屑病表皮细胞过度增殖的调节。Objective To investigate the role of MAPK in the hyp er proliferation of psoriatic keratinocytes.Methods MAPK activity of thirteen ps oriatic epidermis,psoriatic uninvolved epidermis and normal epidermis was assa yed by measuring the transfer of 32 p from γ- 32 P-ATP to my elin basic protein(MBP) and was expressed as pmol/mg protein/min.Results MAPK activity in psoriatic lesions(PL)is 30.46±7.21 pmol/mg/min,that m psoriat ic uninvolved epidermis(PN) is 16.83±3.41pmol/mg/min,that in normal human epidermis(N) is 15.31±3.02 pmol/mg/min.MAPK activity in psoriatic lesion s increased obviously compared with that in psoriatic uninvolved epidermis and normal human epidermis.There were no obvious differences between the MAPK acti vity in psoriatic uninvolved epidermis and that in normal epidermis.Conclusion Hyperproliferation of psoriatic keratinocytes may be closely correlated with h igher activity of MAPK in psoriatic lesions
