肾康片对阿霉素肾病大鼠的肾脏保护作用

摘　　要：目的:探讨肾康片对阿霉素肾病大鼠的肾脏保护作用及其作用机制。方法:经静脉注射阿霉素造成阿 霉素大鼠肾病模型。设立阿霉素肾病大鼠模型组,肾康片高剂量治疗组,肾康片低剂量治疗组,苯那普利治疗组及正 常对照组。6周后观察肾康片对大鼠24h尿蛋白定量、肾功能、血脂及肾脏病理影响,取肾皮质匀浆测超氧化物歧化 酶、丙二醛,应用免疫组化的方法测定肾组织标本纤维连接蛋白、层黏连蛋白、Ⅳ型胶原、转化生长因子-β1、白介 素-1的表达。结果:肾康片可减少阿霉素肾病大鼠的尿蛋白排出量,明显降低甘油三酯,减少纤维连接蛋白、层黏连 蛋白、Ⅳ型胶原、转化生长因子-β1、白介素-1的表达,显著增加超氧化物歧化酶、减少丙二醛,作用与苯那普利相似。 结论:肾康片能减轻阿霉素肾病大鼠的肾脏损害,对肾脏具有保护作用。Objective:To investigate the protective effect of Shenkangpian on adriamycin(ADR) nephrotic rats and the corresponding mechanism.Methods:Nephrotic syndrome was induced by intravenous injection of ADR. Rats were randomly divided into 5 groups:model control group,Shenkangpian high-dosage group ,Shenkangpian low-dosage group, Benazapril group and normal control group. Quantity of 24h urinary protein excretion, albumin, blood cholesterol,triglyceride,serum creatinine,BUN and malondialdehyde(MDA), saproxide dismatase (SOD) and pathological changes of kidney were examined after 6 weeks. Immunohistochemistry was used to examine the expression of fibronectin(FN), laminin(LN): Collagen Ⅳ, transforming growth factor β 1(TGF-β 1) and interleukin one (IL-1) in kidney tissues.Results:The excretion of urinary protein and the triglyceride were significantlly reduced by Shenkangpian after 6 weeks treatment as compared with model group.Immunohistochemistry analysis showed that the expression of FN,LN, collagen Ⅳ, TGF-β 1 and IL-1 in the kidneys were decreased by shenkangpian. MDA in kidney homogenates was decreased,while SOD was increased notably as compared with model group.Conclusion:Shenkangpian can relieve renal leisions of adriamycin nephropathy by decreasing extracellular matrix accumulation, inhabiting TGF-β 1 and IL-1 expression and reducing oxygen free radicals in kidney of rats.
