益寿调脂片对大鼠血糖调节及血液流变学、一氧化氮和内皮素的影响

摘　　要：目的: 探讨益寿调脂片对糖尿病 (diabetesmellitus,DM)防治作用的机制.方法: 利用ip50g/L四氧嘧啶制作DM大鼠模型后,随机分成 5组.分别ig给予10g/L淀粉溶液(空白与模型组)、二甲双胍溶液 (二甲双胍组 )、益寿调脂片溶液(低和高剂治疗组 ). 3wk后观察该药对DM大鼠血糖、血液流变学、一氧化氮 (NO)和血浆内皮素 (ET)的影响.结果: 与模型组比较,该药能够显著增加DM大鼠体质量,具有降低血糖的作用 (P<0 01 ),治疗前后分别为低剂量(22 1±3 1), (17 5±2 1 )和高剂量 (21 6±4 1), (16 1±3 0)mmol/L,与二甲双胍作用相似.并能够显著改善DM大鼠血液流变学(P<0 01 ), ( 6 24±0 67 ), ( 21 10±3 56 )和(1 35±0 08), (1 71±0 18) (低剂量 ) (mPa·s), (4 98±0 94), (17 23±4 06)和(1 33±0 06), (1 63±0 26) (高剂量) (mPa·s).可增加血清NO和降低血浆ET(P<0 01 )水平,低高剂量改变 ( 88 3±6 2 ), ( 97 2±9 2 ) (μmol/L)和(64 5±3 4), (56 7±2 5) (ng/L),且具有一定的量效关系.结论: 益寿调脂片具有一定的降糖作用,其降糖作用的机制可能与其改善血液流变及NO和ET干预有关.AIM: To observe the effects of Yishou Tiaozhi (YSTZT) Tablets' prevention and treatment of diabetes mellitus (DM) in alloxan diabetic rats.METHODS: The model of alloxan diabetic rats was induced by abdomen injection 50 g/L alloxan for 7 d and the model rats were randomly divided into five groups.Control group and model group were given 10 g/L amylum solution,Metformin group was given metformin solution,Yishou Tiaozhi Groups (Low dosage group and High dosage group) were given TSTZT solution.Three weeks later,the limosis blood sugar,hemorheology,nitric oxide and endothelia in alloxan diabetic rats were observed.RESULTS: Compared with model group,YSTZT,like Metformin,could obviously increase the rat's bodymass and lower the level of blood sugar (P<0.01).YSTZT also obviously improved the hemorheology of DM rats,increased the level of serum nitric oxide while reduced the level of plasma endothelia (P<0.01).YSTZT had a dose-effect relationship.CONCLUSION: TSTZT has some function of lowering blood sugar,which may come from the improvement of hemorheology and the interference of nitric oxide and endothelia.
