摘 要:以伏令夏橙长期继代培养的愈伤组织诱导体细胞胚发生。切片观察表明:胚性愈伤组织由胚性和非胚性细胞组成,体细胞胚发生以单细胞外起源方式为主,也可能来自胚性细胞团。胚性细胞分裂产生两个细胞,进而分化发育成多细胞原胚、球形胚、心形胚及子叶胚。次生胚和畸形子叶胚的产生较常见。通过PAS反应发现,胚性细胞和球形胚时期有两次淀粉积累高峰,淀粉的消长与体细胞胚发生的能量供应密切相关。Somatic embryos were induced from long-term sub-cultured callus of Citrus sinensis Osbeck cv. Valencia. The histocytological section and microscopic observation showed that the embryogenic calli were composed of embryogenic and non-embryogenic cells. Somatic embryos were mainly initiated from single cells on the surface of embryogenic calli, and also from embryogenic cell clusters possibly. Then embryogenic cell divided into two cells, which sequentially differentiated through multicell proembryo, globular, heart-shaped and cotyledonary embryo stages. Secondary and abnormal embryos were easily observed. Starch changes were studied by the PAS staining method. Results indicated that there were two peaks of starch accumulation, one was in embryogenic cells, the other was in globular embryos. These results suggested that starch was closely related with energy supply of somatic embryogenesis.
