摘 要:目的 探索瓦斯爆炸伤致肺缺氧后肺组织缺氧诱导因子 1α(HIF 1α)mRNA动态变化规律及其在肺继发性损害发病机制中的作用。方法 建立瓦斯爆炸伤动物实验模型。观察瓦斯爆炸后 30min ,2 ,4 ,8,12 ,2 4 ,4 8h时段肺组织形态学变化 ;用免疫组化SP法进行HIF 1α及半胱氨酸蛋白酶 3(Caspase 3)免疫组化染色 ;用逆转录 -聚合酶链反应 (RT -PCR)方法对肺组织HIF 1αmRNA变化进行半定量检测。结果 HIF 1αmRNA及其蛋白表达明显增加 ,2h达高峰 ,明显高于 30min及 4 8h组 ,对照组无表达 ,P <0 0 1。Caspase 3表达明显增加 ,2h达高峰。 结论 HIF 1α在瓦斯爆炸伤致肺脏缺氧的继发性损害发病机制中起重要作用 ,且与损伤程度密切相关。减少HIF 1α及Caspase 3的过量表达 ,可减轻瓦斯爆炸所致的肺脏损伤 ,减轻肺脏的继发性损害及炎症反应。Objective To explore the dynamic changing regularity of HIF-1α mRNA and its production,and to study the roles they played in the consequent damage in lung anoxic injury caused by gas explosion.Methods animal model of gas explosion injury was created.The lung features were analyzed after fire gas explosion in 30?min,12?h,24?h.Semi-quantitative RT-PCR was used to measure HIF-1α mRNA.Immunohistochem of HIF-1 and Caspase-3 in the l ung tissue was measured.Results The expression of HIF-1α mRNA and its protein rised obviously and peaked at 2 hours.It was higher than that at 30?min and 48 hours.There was not express ion in control group(P<0.01).The expression of Caspase-3 rised obviously and peaked at 2 hours.Conclusion HIF-1α played an important role in lung an oxic injury and the consequent damage caused by gas explosion,and it correlated with the injuring severity.The reduce of overexpression of Caspase-3 and HIF- 1α could alleviate the damage and inflammation reaction in lungs of rats.
