摘  要:目的 :研究三七总皂甙对胎鼠皮层神经干细胞增殖、分化、自我更新及BDNF、bFGF表达的作用。方法 :选取E17胎鼠皮层细胞 ,通过原代细胞培养、传代、单细胞克隆和免疫细胞化学方法。结果 :给药组原代、P1、P2细胞都能够形成较多的神经球 ;给药组P1细胞的单细胞克隆能够形成神经球 ;与对照组比较 ,给药组原代细胞培养 4dNestin、PCNA、Tuj - 1、NF、Vimentin、GFAP、bFGF、BDNF阳性细胞数均明显增多 ,阳性细胞能够形成明显的团状。结论 :体外三七总皂甙可能通过促进胎鼠皮层神经干细胞或前体细胞bFGF、BDNF自身合成增加 ,通过自分泌或旁分泌作用 ,促进其存活、增殖、分化和自我更新。Objective: To study the effects of Panax Notoginseng saponins on their proliferation, differentiation, renewal and expression of bFGF and BDNF of cortical NSCs of embryonic rat. Methods: We used cortical NSCs from E17 embryonic rat., and primary cell culture, passage, monoclone and immunocytochemistry. Results: the primary cells, P1 cells, and P2 cells of treatment group formed neurospheres; Monoclone of the P1 cells of treatment group also formed neurospheres; Compared to control group, the primary cells cultured 4d in vitro of treatment group, had increased numbers of cells that positive to Nestin, PCNA, Tuj-1, NF, Vimentin, GFAP, bFGF, and BDNF. These positive cells formed clusters obviously. Conclusion: These rerults indicated Panax Notoginseng saponins might promote the survival, proliferation, differentiation and self-renewal of NSCs or neural precursors in the cortex of embryonic rat by increasing the synthesis of bFGF and BDNF by themselves.
