摘  要:本文用HPLC柱切换技术,以MercklichroprepRP2(25~40μm)为预处理柱填料,ShimpackCLC-ODS为分析柱,水为预处理流动相,0.2mol/L乙酸和乙酸铵为净化清洗液,在线净化富集样品;通过柱自动切换,以甲醇-乙酸铵(0.2mol/L,pH2.7)(32∶65)为分析流动相,将净化富集于预处理柱上的被测组分冲至分析柱上分离,在280nm处测定。本法操作简单,血浆中环丙氟哌酸含量以外标法定量,日间相对标准偏差为0.7~2.2%,日内相对标准偏差为1.1~3.3%,在浓度为103~5150ng/ml范围内线性良好,r=0.9999,最低检测浓度达52ng/Ml血浆。An automated clean-up and concentration method by column switching is described for the assay of ciprofloxacin, an antimicrobial agent, in plasma. The system uses Merck Lichoprep RP 2 (25~40μm) as a short precolumn packing material for on-line clean-up and concentration. Shimpack CLC-ODS(5μm) is used as analytical colunm. Water is used as the pretreatment mobile phase and a mixture of methanol-0.2mol/L ammonium acetato solution (pH2. 7) (32 ∶ 65) is used as the analytical mobile phase. The plasma is diluted with a solution of 0.2mol/L acetic acid and injected directly on the precolunm. A solution of 0.2mol/L acetic acid and a solution of 0. 2mol/L acetic acid-ammonium acetate are used as the purge solution for clean-up. The detection is at 280 nm and no internal standard is needed.The RSD within day are 1. 1~3. 3% and the RSD between days are 0. 7~2. 2%. In the range of 103~5150 ng/ml plasma the correlation is good with r= 0. 9999. The detection limit is 52 ng/ml plasma.
