摘 要:①目的 探究青紫薯色素对60Coγ 射线辐射损伤小鼠的保护作用及机制。②方法 复制60 Coγ 射线(辐射强度为6 Gy)对雄性昆明种小鼠辐射损伤的病理模型。实验设计分为6组:对照组,60Co模型组,小剂量青紫薯色素组(青紫薯色素15 mg/kg),中剂量青紫薯色素组(青紫薯色素150 mg/kg),大剂量青紫薯色素组(青紫薯色素1 500 mg/kg)和尼尔雌醇阳性对照组(尼尔雌醇2 mg/kg),均灌胃给药,连续1个月。全血细胞分析仪计数小鼠外周血白细胞总数;计算小鼠的胸腺系数和脾脏系数;酶生化法测定小鼠脑、胸腺、肝、脾、肺组织中超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH px)活性,活性氧(ROS)、丙二醛(MDA)含量以及总抗氧化能力(T AOC);油镜下检测小鼠骨髓细胞的微核率和染色体畸变率。③结果 在6 Gy 60Coγ 射线辐射情况下,青紫薯色素(灌胃剂量为15~1 500 mg/kg)能升高小鼠胸腺系数和脾脏系数,显著提高小鼠脑、胸腺、肝、脾、肺组织中SOD和 GSH px活性及T AOC,减少上述组织中ROS、MDA含量(F=14.07~1 315.68,q=5.51~80.49,P<0.01)。青紫薯色素的作用呈剂量依赖性。但青紫薯色素不能抑制辐射所致的小鼠白细胞总数下降、骨髓细胞产生微核及染色体畸变。④结论 青紫薯色素能抑制60Co辐射诱导的小鼠脑、胸腺、肝、脾。Objective To investigate the anti-irradiation effect of Ipomoea Batatas Poir Cv anthocyanins on mice irradiated by 60Co gamma ray in vivo. Methods The irradiation-damage models of Kunming mice were established by 60Co gamma ray(6.0 Gy) in vivo. The animals were randomly divided into six groups, including control group and five 60Co groups (model group,low dose,middle dose,and high dose Ipomoea Batatas Poir Cv anthocyanins groups, and nilestriol positive control group). The spleen and thymus indices were calculated,and the white cells of the animals counted. The activities of superoxide dismutase(SOD),glutathione peroxidase(GSH-px),malondialdehyde(MDA),reactive oxygen species(ROS) and total antioxidative capacity (T-AOC) of the animals were measured. The formation of micronuclei and the mutation of chromosomes in mice were also measured. Results At the dose of 6.0 Gy,Ipomoea Batatas Poir Cv anthocyanins could increase the spleen and thymus indices,inhibit the decrease of SOD,GSH-px and T-AOC activities and the increase of the amount of MDA and ROS of several organs in mice dose-dependently(F=14.07-1 315.68,q=5.51-80.49,P<0.01). But Ipomoea Batatas Poir Cv anthocyanins had no influence on the amount of white cells,the formation of micronuclei and the mutation of chromosomes. Conclusion Ipomoea Batatas Poir Cv anthocyanins has protective effects on mice irradiated by 60Co in vivo. The mechanisms might be attributed to its anti-oxidative capability and the modulation of the immune system.
