多西他赛对结肠癌LoVo细胞增殖的抑制作用

摘　　要：目的　探讨多西他赛治疗结肠癌的作用机制。方法　采用不同浓度的多西他赛处理结肠癌LoVo细胞后 ,应用四唑蓝比色试验检测细胞增殖 ,采用端粒重复扩增 -微孔板杂交法检测端粒酶活性 ,分别用琼脂糖凝胶电泳和TUNEL方法检测细胞凋亡 ,采用流式细胞仪测细胞周期。结果　多西他赛对结肠癌LoVo细胞增殖具有较强的抑制作用。多西他赛能够抑制端粒酶活性 ,可诱导结肠癌细胞凋亡 ,均呈浓度和时间依赖性。结论多西他赛抑制结肠癌细胞恶性增殖与抑制端粒酶活性、诱导凋亡及细胞周期阻滞有关。Purpose To identify the mechanism of docetaxel on colon carcinoma LoVo cell line. Methods After colon cancer LoVo cell were treated with docetaxel,the supression rate,telomerase activity,apoptosis and cell cycle of colon cancer Lovo cell were studied by MTT assay,TRAP-microtiter plate hybridization assay,agarose gel electrophoresis and terminal uridine deoxynucleotidyl nick end labeling(TUNEL),and flow cytometry,respectively. Results Docetaxel had a remarkable inhibition on colon cancer LoVo cell line.It could suppress the telomerase activity,induce cell apoptosis and cancer cells in G 0/G 1 phase arrest.The effect of supression of telomerase activity and induction of apoptosis were relevant to the time and density of docetaxel. Conclusions Docetaxel can inhibit the growth of huamn colon cancer LoVo cell by inhibiting the expression of telomerase and inducing apoptosis and G 0/G 1 arrest.
