摘 要:目的 观察糖尿病多发性神经病 (DPN)大鼠坐骨神经NGF的动态表达以及胰岛素和人神经生长因子(hNGF)的干预作用。方法 以链脲佐菌素 (STZ)诱导糖尿病大鼠模型 ,通过RT PCR和免疫组化技术 ,观察造模后 4和 8周大鼠坐骨神经NGFmRNA和蛋白的表达 ,并观察DPN 4周时用胰岛素或hNGF干预 4周后坐骨神经NGFmRNA和蛋白的表达。结果 NGFmRNA表达在DPN 4周无明显改变 ,8周时明显减少 ;NGF蛋白表达在DPN 4周和 8周均减少。hNGF干预能增加DPN大鼠坐骨神经NGFmRNA和蛋白表达 ,胰岛素则无明显调节作用。结论 DPN大鼠坐骨神经NGF表达减少 ,外源性NGF可弥补这种缺陷。Purpose To observe the of nerve growth factor(NGF) in sciatic nerves of diabetic polyneuropathy(DPN) rats,and the effect of insulin or human nerve growth factor. Methods Diabetic rats were induced by single intraperitoneal injection of Streptozotocin (STZ).The expressions of NGF mRNA and protein in sciatic nerves of DPN 4 wk or 8 wk rats were observed by RT-PCR and immuneohistochemistry.The effects of insulin or human nerve growth factor on NGF mRNA and protein in sciatic nerves were also observed by same techniques. Results NGF mRNA expression was not reduced significantly until DPN 8 wk.NGF protein were decreased significantly at both DPN 4 wk and 8 wk.After treatment with hNGF for four weeks,both NGF mRNA and protein expressions were increased significantly.But insulin had no effects on them. Conclusions NGF expressions were decreased in the sciatic nerves of DPN rats. Exogenous NGF may approve the deficits.
