摘　　要：利用生物信息学方法成功地克隆了家蚕磷酸甘油醛异构酶 (triosephosphate isomerase Tpi) 基因 (GenBank登记号为AY734490).该基因全长为2 875 bp;含有5 个外显子、4个内含子,所有内含子/外显子边界都符合典型的GT/AG剪切模式;cDNA 序列全长1 051 bp,并经RT-PCR克隆和序列分析进行了验证;具有完整的开放阅读框架(ORF,95～839 bp),可编码248个氨基酸的蛋白.通过与NCBI蛋白数据库的比对表明,该基因编码的蛋白为磷酸甘油醛异构酶(EC 5.3.1.1),参与葡萄糖分解代谢,即将磷酸二羟丙酮转化为3-磷酸甘油醛,在果蝇、蚊子、黄粉虫、美洲淤夜蛾、棉铃虫等昆虫中具有高度的保守性.A novel silkworm,Bombyx mori,triosephosphate isomerase(Tpi) gene is cloned by blasting its EST database and PCR test,and its sequence has been deposited in the GenBank (Accession No: AY734490).It has five extrons and four introns spanning 2 875 bp in full length and all the boundary of extrons/introns has GT/AG conserved sequences.Its 1 051bp mRNA has a complete open reading fragment (ORF: 95-834) and encodes 248 Aa protein,which is triosephosphate isomerase(EC 5.3.1.1)for catalysis of conversion dihydroxyacetone phosphate into glyceraldehydes 3-phosphate in glucose glycolysis and very conserved in many insects such as Heliothis virescens,Helicoverpa armigera,Tenebrio molito,Anopheles gambiae and Drsophila melanogaster.
