摘  要:目的观察实验性糖尿病大鼠海马突触超微结构改变及胰岛素治疗的作用。方法将成年SD大鼠随机分为正常对照组、糖尿病组、糖尿病治疗组,应用透射电镜和形态计量学方法观察分析成模第3月末及第6月末海马突触超微结构及结构参数的变化。结果糖尿病组可见突触变性,突触密度、突触后致密物厚度显著减低,突触间隙宽度显著增宽。6月组较3月组更加严重。治疗组仅6月时有显著改变,但较糖尿病6月组明显轻微。结论糖尿病可导致大鼠海马突触数量减少、形态结构发生显著变化,早期应用胰岛素治疗可预防、延缓病变。Objective To study the ultrastructural changes in hippocampus' synapses of diabetic rats and the effect of insulin on these changes. Methods Adult Sprague-Dawley rats(BW 250g±) were divided into normal control group, diabetic group and treatment group. Rats were killed at 3 months and 6 months after Morris water maze test.The Ultrastructural changes of synapses were observed in electron microscope. The number density,the width of synaptic cleft and the thickness of postsynaptic density were studied. Compared to the normal control group,synapses of diabetic group had retrograde changes. Results The number of density, the thickness of postsynaptic density were decreased and the width of synaptic cleft were significantly increased at 3 months(P<0.01), and more significantly at 6 months(P<0.01). In treated group only 6 months groups had such changes. Conclusion There are significant changes in hippocampus' synapses of diabetic rats and early use of insulin could delay the occurrence of neuropathological changes.
