摘  要:以辣椒属长椒变种的两个自交系杂交所得的F1代经自交获得F2 代的 93个单株为作图群体 ,利用RAPD技术开展了辣椒分子遗传图谱的构建研究 :选用Operon公司的 3 2 0个随机引物对亲本进行了RAPD多态性检测 ,通过多次重复筛选 ,获得了 3 3个多态性引物 ,得到 49个具有较好重复性和稳定性的RAPD标记 ,其中 11个标记表现为偏分离标记 ;利用MAPMAKER/expversion 3 .0软件初步构建了辣椒的分子连锁图谱 ,该图谱由 11个连锁群组成 ,含有 2 8个RAPD标记 ,总长度为 2 82 .41cm。In this study, RAPD Markers were employed for construction of a linkage map using 93 plants of F_2 population. The F_2 mapping population was generated by selfing a single F_1 plant obtained from the cross between the two inbred lines of capsicum annuum L. var. grossum. A total of 320 random primers were used to detected DNA polymorphism between the parents. Through several repeatable assays, only 33 of these random primers used were useful in terms of polymorphism detected and the reproducibility and stability of those patterns , and 49 polymorphic segregating loci were identified. Of those, only 11 markers showed distorted segregation. A preliminary molecular linkage map has been constructed by Mapmaker/exp version 3.0 software. 28 linkage markers were assigned to 11 linkage groups covering a total length of around 282.41 cm.
