摘 要:从浙江丽水的亚洲百合病叶中得到两病毒分离物G和X,电镜观察病组织汁液中含有大量线状病毒粒子,病叶细胞质中存在着柱状内含体.按照Potyvirus成员和百合斑驳病毒(Lilymottlevirus,LMoV)的已知基因序列设计特异性引物,经RT-PCR扩增分别得到1692nt和1469nt两个片段,包含NIb、CP和3'-URT,3'-端含poly(A).将CP基因序列与侵染百合和郁金香的10个Potyvirus分离物进行同源性比较,结果表明G、X与LMoV的同源性分别为85.9%~99.4%、86.1%~99.2%(核苷酸)和87.0%~96.7%、89.9%~97.8%(氨基酸),两分离物均为LMoV.系统进化树分析显示G和X分别处在LMoV的两个群体中,同源性较低,不表现地域相关性及寄主相关性.Two isolates, G and X were gained from Lilium cvs. (Asiatic hybrids) in Lishui of Zhejiang Province. Numerous filamentous virus particles resembling Potyvirus were detected by electron microscopy. Pinwheel inclusions were observed in the cytoplasm of parenchyma cells. Based on a primer specially designed for the genome sequences by the member of Potyvirus and Lily mottle virus (LMoV), two partial sequences of cDNA were cloned by RT-PCR. They were composed of 1692 nt and 1469 nt respectively, both including NIb, CP, 3'-URT, and poly(A) of 3′ end. Similarity of CP sequences between G and X isolates were compared with ten isolates of Potyvirus infecting lily and tulip plants from other region of the world. The result showed that G and X have high homology with LMoV in CP nucleotide sequence and CP amino acid sequence. They varied from 85.9% to 99.4%, 86.1% to 99.2% and from 87.0% to 96.7%, 89.9% to 97.8% respectively, and the two isolates was thus confirmed as LMoV. The phylogenetic tree topology analysis suggested G and X be divided into two different groups. They have low homology and their evolution were not related with the host and geological origination.
