薰洗方对大鼠骨折后期僵硬关节滑膜Ⅲ型胶原mRNA及MMP1活性的影响  被引量:3

摘  要:目的:探讨薰洗方治疗骨折后期关节僵硬的作用机理。方法:以Wistar大鼠制作骨折后关节僵硬模型,用dotblot杂交法和Maruyama法分别测定踝关节滑膜 型胶原m RNA水平和MMP1活性。结果:模型组 型胶原m RNA水平明显升高,MMP1活性明显增强,与正常对照组比较差异有显著性(P<0 .0 1 ) ;功能组 型胶原m RNA水平显著低于模型组(P<0 .0 1 ) ,而MMP1活性略有降低,与模型组比较差异无显著性(P>0 .0 5 ) ;中药组 型胶原m RNA水平、MMP1活性明显降低,与模型组比较差异有显著性(P<0 .0 1 ) ;同时中药组 型胶原m RNA水平、MMP1活性显著低于功能组(P<0 .0 5 )。结论:大鼠骨折后期僵硬关节滑膜 型胶原m RNA表达及MMP1活性异常改变;降低 型胶原m RNA表达水平及MMP1活性可能是薰洗方治疗骨折后期关节僵硬的重要机制之一。Objective:To investigate treating mechanism of XunXinFang on joint stiffness of rats in the late-stage of fracture.Methods:Wistar rats were taken as animal model of joint stiffness in the late-stage of fracture . Dot blot hybradizition and Maruyama method were respectively used to measure the level of type Ⅲ collagen mRNA and matrix metalloproteinase-1(MMP1) activities in synovium of stiff ankle joint of rats .Results: the level of type Ⅲ collagen mRNA and MMP1 activities significantly increased in the model group and the difference was significant compared with the normal control group (P<0.01).The level of type Ⅲ collagen mRNA in the function group was obviously higher than that in the model group(P<0.01). MMP1 activities in the function group decreased,but the difference was not significant compared with the model group(P>0.05).The level of type Ⅲ collagen mRNA and MMP1 activities in the Traditional Chinese Medicine group obviously decreased and the difference was significant compared with the model group(P<0.01).At the same time ,the level of type Ⅲ collagen mRNA and MMP1 activities in the Traditional Chinese Medicine group obviously eleveted compared with the function group (P<0.05).Results:Abnormal changes of the level of type Ⅲ collagen mRNA and MMP-1 activities exist in synovium of stiff ankle joint of rats.Decreasing the expression of type Ⅲ collagen mRNA and MMP-1 activities may be one of important treating mechanisms of joint stiffness in the late-stage of fracture with XunXinFang.
