摘  要:目的研究半边旗二萜类化合物5F(11α-羟基-15-氧-16-烯-对映贝克杉烷-19酸)对人高转移卵巢癌细胞系HO-8910PM细胞内核转录因子κBP65亚基[NF-κB(P65)]、Smad3蛋白及NF-κB(P65)、ETS-1mRNA表达的影响,探讨其抗肿瘤的作用机制。方法MTT法检测5F对HO-8910PM细胞增殖的影响;Westernblot分析NF-κB(P65)及Smad3蛋白的表达;RT-PCR分析NF-κB(P65)及ETS-1mRNA的表达。结果5F能抑制HO-8910PM细胞的增殖,抑制率随剂量的增加而增加,具有剂量依赖性;25~100μmol/L5F作用HO-8910PM细胞24h后,NF-κB(P65)、Smad3蛋白表达水平明显下降,并呈剂量效应关系;5F明显下调NF-κB(P65)mRNA的表达,轻微下调ETS-1mRNA的表达。结论5F抗肿瘤活性与NF-κB(P65)、Smad3蛋白和NF-κB(P65)、ETS-1mRNA的表达有关。Objective To study the effect of a diterpenoid 5F (ent-11α-hydroxy-15-oxo-kaur-16-en-19-oic acid) isolated from Pteris semipinnata on the expressions of NF-κB (P65), Smad3 protein and NF-κB (P65), ETS-1 mRNA, and to investigate the antitumor mechanisms of 5F. Methods MTT assay was used to examine the effect of 5F on proliferation of HO-8910PM cells. The expressions levels of NF-κB (P65) and Smad3 proteins were assessed by Western blot analysis. RT-PCR assay was used to assess the expression levels of NF-κB (P65) and ETS-1 mRNA. Results The proliferation of HO-8910PM cells can be inhibited by 5F in a dose-dependent manner. The expression levels of NF-κB (P65) and Smad3 proteins decreased obviously in HO-8910PM cells after treatment with 25—100 μmol/L 5F for 24 h, and the effects appeared in a dose-dependent manner. 5F down-regulated significantly the expression of NF-κB (P65) mRNA, down-regulated gently the expression of ETS-1 mRNA. Conclusion The antitumor activity of 5F is involved in the expression of NF-κB (P65), Smad3 protein and NF-κB (P65), ETS-1 mRNA.
