偏光图像法分析高血压大鼠的心肌胶原重构

摘　　要：目的　通过改进苦味酸-天狼猩红染色心肌胶原偏光图像的分析方法,对高血压大鼠心肌胶原重构程度和药物对重构的改善效果进行评估。方法　建立高血压模型大鼠,取心肌进行特异性的苦味酸-天狼猩红胶原染色,在正交偏光下采用不同角度观察I、III型胶原的染色情况,分析偏光显微镜观察方式对胶原成像影响的物理机制并提出图像采集方式上的改进。采集图像后,用迭代阈值法获取阈值对胶原图像进行自动分割,对分割出的不同胶原成分计算形态学指标。同时考察正常组与治疗组的相应指标。结果　胶原面积和容积分数等指标表明高血压大鼠心肌胶原含量明显高于正常组,I、III型胶原成分也明显和正常组不同。治疗组较高血压组与正常组接近。结论　通过对心肌胶原进行苦味酸-天狼猩红染色,利用偏光显微镜,结合图像分析技术,选择合适的指标,能够对高血压引发的心肌胶原重构进行较为有效的量化评估。OBJECTIVE An improved technique based on picro-siriusred polarization and image analysis was investigated for evaluation of collagens remodeling in myocardium of hypertensive rats. METHODS The myocardium tissue sections of hypertensive rats were stained with picro-siriusred. The content and distribution of collagens were observed with polarization microscopy, and then rotated sections, took 2 pictures spacing 45 degree by camera and did image analysis. The collagens were segmented with image analysis system and different types were distinguished by color values. Interstitial collagen volume fraction and the ratio of I type collagen to III type collagens were measured. RESULTS In the view of polarization microscopy, I type was red and III type was green. The intensity of color was changing with rotating of section, and came back to original intensity after 90 degree. The image analysis showed apparenty differences between hypertensive rats and control rats. the ratio of I type to III type was also different between hypertensive rats and treated rats. CONCLUSION This study demonstrates that the usefulness of improved picro-siriusred polarization method and image analysis as an effective tool for detection different type of collagen in studying of myocardium remodeling.
