摘 要:以鲁梅克斯K-1杂交酸模的幼嫩花序切段为外植体,在诱导培养基上经连续4代培养,诱导丛生芽高频率发生.丛生芽增生迅速,在继代培养中增殖率达每月10~40倍.丛生芽分割为单芽后,转入生根培养基中产生根系.旺盛增生的丛生芽在附加171 mmol/L NaCl的培养基上连续筛选3代,获得了耐盐小芽,然后在无NaCl的培养基上形成完整植株.耐盐小芽在无NaCl的培养基上培养3代后转回附加171 mmol/L NaCl的培养基上继续培养,表现出高的耐盐性和小芽增殖率.耐盐再生植株的后代在盐碱地种植,表现出良好的耐盐性.本研究建立了一个适合于离体遗传操作的鲁梅克斯丛生芽诱导及继代培养体系,并获得了鲁梅克斯耐盐育种新材料,为培育出适合于滨海盐碱地及大面积盐荒地种植的鲁梅克斯新品种奠定了基础.Multiple bud clumps were induced in high frequency from segments of immature inflorescences of Rumex (R. patientia×R. tianschanicus) K-1 on MS medium supplemented with 1 mg/L 6-BA after 4 times of subculture. The bud clumps proliferated vigorously on the same medium and the proliferating rate was almost 10-40 times/month. Single bud was cut down from the bud clumps and was transferred to root-inducing medium containing 1 mg/L NAA to develop to plantlets. Vigorously proliferating buds were subcultured on medium supplemented with 171 mmol/L NaCl for 3 consecutive generations and salt-tolerant buds were obtained. Salt-tolerant buds, after being subcultured for 3 generations on NaCl-free medium, showed higher salt-tolerance and proliferating rate than the control on medium supplemented with 171 mmol/L NaCl. The progenies of the salt-tolerant buds sown in saline soil where the salt concentration was about 0.7%, exhibited better salt-tolerance than the control, but the growth and biomass were less than those in normal soil. These results showed that we had established an efficient multiple bud clumps propagation system of Rumex, which was suitable for the genetic manipulation in vitro. Through this experiment, we also obtained some useful materials of salt-tolerance for Rumex breeding.
