摘　　要：目的　研究D型CpG 寡脱氧核苷酸(CpG- ODN)体外对慢性乙型肝炎患者的免疫刺激作用。方法　分离患者外周血单个核细胞(PBMC), CpG- ODN单独或联合重组人粒细胞巨噬细胞集落刺激因子(rhGM -CSF)刺激培养2天后,用ELISA法检测培养液中IFN α的水平,RT- PCR法检测PBMC中IFN- γ的表达水平。结果　CpG- ODN可有效诱导慢性乙型肝炎患者PBMC分泌IFN -α,高表达IFN -γmRNA。结论　CpG -ODN对慢性乙型肝炎患者有明显的激活作用。Objective To investigate the effect of phosphorothioate oligodeoxynucleotides (ODN) containing unmethylated CpG dinucleotides (CpG-ODN) on peripheral blood mononuclear cells (PBMCs) from the patients with chronic hepatitis B.Methods CpG-ODN alone or combined with recombinant human granulocyte-macrophage colony stimulating factor (rhGM-CSF) was co-cultured with PBMCs.IFN-αlevel in the cultural supernatant was measured by ELISA;the reverse transcription PCR was used to analyze the expression levels of IFN-γ mRNA in PBMCs.Results CpG-ODN induced high secretion of IFN-αand increased mRNA expression of IFN-γin PBMC.Conclusion D type CpG-ODN displays a strong activation in the immune response of the patients with chronic hepatitis B.
