摘 要:目的 观察碱性成纤维细胞生长因子(bFGF)在体内是否具有诱导骨髓间充质干细胞(MSCs)向皮肤组织分化的作用及其在皮肤创面愈合中的作用。 方法 抽取小型猪骨髓,体外分离、纯化、培养MSCs,并用5-溴脱氧尿嘧啶(BrdU)进行标记。6只小型猪共做48个创面,随机分为bFGF+MSCs治疗组(A组)、MSCs治疗组(B组)、bFGF治疗组(C组)和等渗盐水对照组(D组),观察伤后愈合情况。在伤后3, 7, 14, 21, 42d取标本,并用病理学、免疫组化方法对创面的修复质量进行评估。 结果 A组的皮肤创面愈合速度明显快于B、C和D组;A组在皮肤愈合的组织病理等级评分上优于B、C组;A组双染阳性细胞多于B组。 结论 在皮肤修复过程中,bFGF具有促进MSCs向血管内皮细胞分化,促进皮肤愈合的作用。Objective To investigate the role of basic fibroblast growth factor (bFGF) in inducing the differentiation of mesenchymal stem cells (MSCs) to skin tissues in vivo and discuss its effect on skin wound healing. Methods MSCs were isolated from porcine marrow, purified and cultured with 5-bromodeoxyuridine (5-BrdU) as a label in vitro. Forty-eight skin wounds on the six porcines were divided into bFGF+MSCs (Group A), MSCs (Group B), bFGF (Group C) and control group randomly and treated correspondingly. Wound closing index (WCI), cell morphology and histological changes in the defected regions were evaluated 3,7,14,21 and 42 days after treatment. Vessel endothelium cells derived from MSCs in the granulation tissues were assayed by immuno-staining of 5-BrdU and Factor Ⅷ in the same cell. Results Healing speed of defected regions in Group A was faster than that of the other groups. Hierarchical scores of histopathology on the specimens of Group A were also higher than that of the other groups. The dual-positive stained cells by BrdU and Factor Ⅷ in Group A were more than those in Group B. Conclusion bFGF can induce MSCs differentiating into vessel endothelium cell and hence promote the repair of skin wounds.
