摘  要:目的:探讨脑缺血后单核巨噬细胞浸润情况。方法:用免疫组织化学方法检测了30只大鼠大脑中动脉局部脑缺血1 2 h、2 4 h、3d、7d和1 0 d后损伤区ED2 阳性巨噬细胞浸润情况。结果:缺血1 2 h脑实质内可见ED2 阳性巨噬细胞,主要分布于坏死区周围;缺血2 4 h,阳性细胞较1 2 h有所增加,室管膜上有大量的阳性细胞聚集;缺血3d时阳性细胞数达高峰,主要存在于损伤区的周边区。缺血7d、1 0 d时染色中仅偶见阳性细胞的存在。统计学分析大鼠脑缺血1 2 h、2 4 h、3d时ED2 阳性巨噬细胞与正常对照组有非常显著性差异( P<0 .0 1 ) ,缺血3d组与1 2 h组相比差异也有显著意义( P<0 .0 5 )。结论:巨噬细胞参与了缺血性脑损伤的病理过程。Objective: To explore macrophage infiltration in focal cerebral ischemia of rat. Methods: The ED_2 positive macrophage was observed with the immunohistochemistry.The rats were subjected to middle cerebral artery occlusion(MCAO) for 12h,24h,3d,7d, and 10d. Results: The ED_2 positive macrophage was seen at hour 12 after MCAO around infarct area. At hour 24, there were more ED_2 positive macrophage than at hour 12 after MCAO , many macrophages appeared in ependyma. The cell number was the most at day 3 around infarct area. A few ED_2 positive macrophage were present in the cerebral tissue at day 7 and day 10. The cell number in the cerebral tissue at hour 12, hour 24 and day 3 Was significantly higher in comparison with controls( P <0.01),and there was also remarkable difference between day 3 and hour 12( P <0.05). Conclusion: Macrophage may be associated with the ischemic injury of brain , mainly in the early time of cerebral ischemia.
