摘 要:目的比较不同因素对气道黏液高分泌的诱导作用,为动物模型制造和临床防治提供指导。方法Wistar大鼠30只随机分成3组:正常组雾化吸入生理盐水;对照组雾化吸入脂多糖(LPS);试验组雾化吸入中性粒细胞弹性蛋白酶(NE)。采用酶联免疫吸附试验(ELISA)、免疫组织化学和斑点印迹法分别测定支气管肺泡灌洗液中黏蛋白(MUC)的含量、气道上皮MUC5AC以及MUC5AC mRNA表达。结果试验组和对照组比正常组的MUC,MUC 5AC以及MUC5AC mRNA水平均显著增加(P<0.01);试验组MUC,MUC5AC以及MUC5AC mRNA表达较对照组明显增加(P<0.05)。结论LPS和NE均为黏液高分泌的有效诱导因素,但NE的黏液促分泌效应较LPS更显著,为强有力的诱导剂,是理想的黏液高分泌造模因子,也是临床防治的重要靶目标。Objective To compare the inducing effects of different secretagogues on hypersecretion of respiratory mucosa. Methods Thirty Wistar rats were randomly divided into 3 groups,ie, normal group, control group and trial group;they were put into lucite bins and inhaled saline(0.9%NS), lipopolysaccharide(LPS)and neutrophil elastase (NE) respectively. The levels of mucins (MUC) in bronchoalveolar lavage fluid (BALF),MUC5AC in airway epithelia and MUC5AC mRNA in lung tissues were determined using ELISA, immunohistochemistry and dot blot assay respectively. Results The content of MUC, the positive stain area of MUC5AC and the express of MUC5AC mRNA in the trial group and control group were significantly increased in comparison with the normal group(P <0.01), while the three above-mentioned items in trial group were apparently increased in comparison with the control group(P <0.05). Conclusion LPS and NE are mucus secretagogues. NE has stronger inducing effect on mucus secretion than LPS, it is a potent secretagogue and an ideal agent for setting up respiratory mucous hypersecretion model.NE is also a important target of prevention and cure in the clinic.
