摘  要:目的 检测α-MSH在瘢痕疙瘩成纤维细胞中的表达和作用,为研究瘢痕疙瘩的形成机理提供新的理论依据。方法 将取自患者躯干部位的瘢痕疙瘩皮肤,用组织块培养法进行成纤维细胞原代培养,DMEM培养液传代、扩增培养,以细胞形态学鉴定成纤维细胞。应用免疫组织化学染色方法检测α-MSH在瘢痕疙瘩成纤维细胞中的表达,并应用流式细胞仪从细胞生长及增殖特性方面分析α-MSH对瘢痕疙瘩成纤维细胞生长和增殖的影响。结果 α-MSH能够在瘢痕疙瘩成纤维细胞中得到高表达,浓度为10 -6mmol/L的α-MSH可明显促进瘢痕疙瘩成纤维细胞生长、增殖。结论 α-MSH能够在瘢痕疙瘩成纤维细胞中得到高表达,瘢痕疙瘩成纤维细胞对α-MSH的反应性增强表明α-MSH可能在促进瘢痕疙瘩的形成中起着重要的作用。Objective To study the expression and effects of α-melanocyte-stimulating hormone(α-MSH) in fibroblasts from keloids for finding a novel etiology of keloids. Methods Fibroblasts isolated from human keloids, hypertrophic scar and normal skin tissues were cultured in DMEM medium with 15% bovine serum. The cells were identified by cellular morphology. The expression of α-MSH in keloid-derived fibroblasts was detected by immunohistochemistry. The fibroblasts were then treated with α-MSH at the concentration of 10 -6 mmol/L. The effects of α-MSH on the distribution of cell cycle of fibroblasts were analyzed with flow cytometry. Results The expression of α-MSH could be detected in keloid-derived fibroblasts, and α-MSH could promote the growth and proliferation of keloid-derived fibroblasts. Conclusion The high expression of α-MSH in keloid-derived fibroblasts suggest that α-MSH may play an important role in the keloid formation.
