摘 要:目的 观察链脲佐菌素(STZ)糖尿病大鼠下颌下腺超微结构变化。方法 SD雄性大鼠18只,随机分为对照组、实验组及治疗组,用STZ复制糖尿病动物模型,饲养12周后测体重和血糖,取材制片应用透射电镜观察其超微结构变化。结果 与对照组体重[ ( 367 .33±26. 73 )g]和血糖[ (6 .49±0 .79 ) mmol/L]比较:实验组体重[ ( 156. 50±10 .13)g]减轻、血糖[ (27 .79±9 .28)mmol/L]升高,有显著性差异(P<0 .01);治疗组体重[ (354 .67±31 .94)g]、血糖[ (8. 75±2 .56)mmol/L]无显著性差异(P>0 .05)。与对照组比较,实验组下颌下腺电镜下主要变化为为:细胞核固缩,染色质致密;线粒体肿胀或空泡变性、嵴断裂;粗面内质网囊状扩张,断裂成短棒状,脱颗粒等。治疗组无明显改变。结论 糖尿病可导致下颌下腺组织出现一系列超微病理改变,为进一步探讨糖尿病发生机制提供了形态学依据。Objective To observe the ultrastructure changes in submandibular glands of diabetes rats. Methods Sprague-Dawley rats were randomly divided into control group,experiment group and treatment group, 6 rats per group. The diabetes rats model was induced by injection of streptozotocin into peritoneal cavity. After 12 weeks, the rats were killed for assaying blood sugar and weight ,and the submandibular glands ultrastructure was observed with transmission electron microscope(TEM). Results Blood sugar of the experiment group was (27.79±9.28) mmol/L; and the control group, (7.23±0.84) mmol/L. Weight of the experiment group was (156.50±10.13) g; and the control group, (305.50±34.67) g, the differences were significant ( P <0.01). Under TEM the ultrastructures of rats’ submandibular glands in contol group were normal. While in experiment group a series of pathologic changes were found. The cell nucleus mainly appeared karyopyknosis. Most mitochondria exhibited swelling,vacuole degeneration and cristac breaking. Rough endoplasmic reticula(RER) expanded,the cisternac of the RER were distented and showed degranulation.The ultrastructures of treatment group were similar to control group. Conclusion A series of pathologic changes in submandibular glands of diabetes rats can be induced by diabetes,which will provide morphological evidences for further investigation of mechanism of the diabetic complication.
