摘 要:目的 建立大鼠胚脑皮层神经干细胞体外培养及鉴定技术,探讨其增殖分化特性,为应用神经干细胞移植治疗神经疾病提供基础。方法 采用含有丝分裂源表皮生长因子(EGF)的无血清培养基培养SD大鼠孕14 5d的胚胎大脑皮层神经干细胞,应用免疫细胞化学方法了解其增殖分化特性; 以单细胞克隆实验及BrdU免疫标记证实神经干细胞自我增殖能力。结果 培养细胞在EGF作用下可分裂增殖,并表达神经干细胞特异性抗原nestin,分化细胞MAP2 及GFAP抗原表达阳性。结论 SD大鼠胚脑皮层可分离培养出神经干细胞;并具有增殖、自我更新能力和多向分化潜能特性。Objective To establish the culture and identification methods of cortex-originated neural stem cells from embryonic rat brain,and study their characterization of differentiation and proliferation, in order to provide a cell model for studying neural stem cells. Methods The rat embryonic E14.5 neural stem cells were isolated and cultured in serum-free medium containing the mitogen epidermal growth factor(EGF) and B27. The characterization of neural cells was identified by immunocytochemistry. Their self-renewal capacity was showed by single-cell clonal analyses and BrdU immunocytochemistry. Results The neural stem cells could be propagated extensively in culture medium containing EGF, and were positively immunoreactive for neural stem cells marker nestin. While induced to differentiate into neuronal-like cells, they were showed to express neuronal cell markers MAP 2 or GFAP. Conclusion Neural stem cells can be isolated and cultured from rat embryoric brain cortex.The cultured cells preserve critical characteristics of neural stem cell, including proliferation, self-renewal and multipotency.
