摘  要:目的 确定蓝氏贾第虫病毒体外转录体电穿孔转染的最佳条件。方法 以不同的电击缓冲液、电压及脉冲时间应用GLV和GFP嵌合体体外转录体电穿孔转染蓝氏贾第虫,测定转染虫体的存活率及GFP表达量。结果 在不同转染条件下GLV和GFP嵌合体体外转录体均能成功转染蓝氏贾第虫,且在cytomix缓冲液、10 0 0V/cm、8ms电击条件下,对虫体损伤最小,GFP表达量最高。结论 本试验确定的蓝氏贾第虫病毒体外转录体电穿孔转染最佳条件是电击缓冲液为cytomix缓冲液,电压为10 0 0V/cm、脉冲时间为8ms。To determine the optimal transfection condition for the in vitro transcripts from Giardia lamblia virus (GLV), the transcript from chimera of GLV and the green fluorescent protein (GFP) was electroporated into Giardia lamblia in different buffers, voltages and impulse times. The survival rate of parasites was calculated, and the GFP expression was determined by ELISA. Experimental result showed that the transcript from chimera of GLV and GFP was successfully transfected into Giardia lamblia. The damages on the parasites was minimized and the expression of GFP was to be highest when the optimal conditions for transfection was established in cytomix buffer with the electroporation under 1000 V/cm and 8ms.
