甜橙汁囊培养的脱分和再分化

摘  要:以雪柑(甜橙,CitrussinensisOsbeck)果实不同发育时期的汁囊为外植体,在添加不同激素的MS培养基中,均脱分化获得了愈伤组织,但衰老汁囊获得率低。在培养过程中,接种后10天内可溶性固形物和含酸量迅速下降,10天后又趋于平稳。同工酶凝胶电泳分析表明:成熟汁囊未见水溶性蛋白质带和过氧化物酶带,但经过15天培养,开始产生愈伤组织,谱带明显增加,随培养时间增加,谱带加强,汁囊愈伤组织培养165天后,分化出了根、胚状体和类汁囊,类汁囊具汁囊相似的形态解剖特征。The juice sac explants at different development stage of Xue Gan aweet orange,(Cit-rus sinensis Osbeck)fruits dedifferentiated and produced calli on MS with different hormones. However,the senescent get low production. During the training,soluble solids and acid content de-creased rapidly 10 days after nioculation and tended to be stable 10 days latwr. The result from isozyme PAGE showed tnat the call neither soluble protein bands nor peroxidase bands appeared in adult sac sample,but they were traced after 15-day′s training,and got stronger afterwards. Laterafter 165-day′s training,root embryoid and thylakoid differentiated and the last one was same in morphogenesis and anatomy as juice sac, The result provided a basis for seedling induction of juice sacs.
