摘  要:目的 检测诊断超声辐射与人早孕绒毛Caspase- 3、8蛋白表达的关系,探讨诊断超声诱导滋养层细胞凋亡增加的机制。方法 将拟行人工流产的2 4例早孕(停经4 0~6 0 d)妇女随机分为4组:对照组、1 0 min组、2 0 min组、3 0 min组。经腹部B超对孕囊进行持续照射,2 4 h后取材。用免疫组织化学技术检测各组绒毛滋养层细胞Caspase- 3、8蛋白表达情况。结果 1 0 min组绒毛滋养层细胞Caspase- 3、8蛋白表达率与对照组无差别( P>0 .0 5 ) ;2 0 min组和3 0 m in组绒毛滋养层细胞Caspase- 3、8蛋白表达率明显大于对照组和1 0 min组( P<0 .0 5 ) ,并随辐射时间的延长而表达增强。结论 诊断超声持续照射早孕孕囊1 0 min以上可引起绒毛滋养层细胞Caspase- 3、8蛋白表达率增加。Objective To investigate the relationship between Caspase-3 and Caspase-8 protein expression in the first trimester pregnant trophoblast and diagnostic ultrasound irradiation,and to preliminary study the mechanism of trophoblasts cell apoptosis induced by diagnostic ultrasound.Methods Twenty-four first-trimester pregnancy women were randomly devided into four groups according to different irradiating time and they were exposed to diagnostic ultrasound for 0,10,20,or 30 min.The chorionic villi were obtained 24 hours after exposure and expression of Caspase-3 and Caspase-8 protein was analyzed by immunohistochemistry.Results In contrast with the 0- and 10-min groups,the rates of expression of Caspase-3 and Caspase-8 protein significantly increased in 20 and 30 min groups(P<0.05).There was no difference between 0-min group and 10 min group.Conclusions Continuous irradiation over 10 minutes by diagnostic ultrasound may result in higher expression of Caspase-3 and Caspase-8 protein in the first trimester pregnancy and may further induce trophoblasts cell apoptosis via extrinsic death pathway.
