B细胞系特异性单克隆抗体ZCH－4－2E8的制备鉴定及其初步应用

摘　　要：本文报告一种识别Ｂ细胞系白血病相关抗原的鼠单克隆抗体ＺＣＨ－４２Ｅ８（ＩｇＧ１），免疫印迹试验示其所识别的抗原分子量为２４５ｋｄ。经多种细胞系、正常外周血细胞成分、骨髓单个核细胞、胚胎肝、脾、胸腺和１３８例急、慢性白血病细胞的鉴定结果表明，２Ｅ８为Ｂ细胞系特异性单抗，其反应谱与ＣＤ１９类似，但其尚能识别早至１８周龄的胚胎脾（Ｂ）细胞，阳性率为３１．９％（ＣＤ１９为０％）。作者认为：２Ｅ８是一种能用于分化早期Ｂ细胞系急性淋巴细胞白血病（ＡＬＬ）的诊断和急性未分化型白血病（ＡＵＬ）鉴别诊断的Ｂ细胞系特异性单抗。A murine monoclonal antibody(McAb)named ZCH-4-2E8(IgG1)able to recognize human B cell related antigen is submitted in this report. The immunoblot assay demonstrated that it recognized an antigen with a molecular weight of 245 kd.Cells from several cell lines,normal peripheral blood,bone marrow, fetal liver,spleen and thymus as well as those from 138 cases of both acute and chronic leukemia were tested with 2E8 to show that 2E8 to be a B lineage specific McAb with a similar spectrum to that of CD19. But the former could also recognize immature B cells as early as in the fetal spleen of 18-week gestational age with a positive rate of 31.9%but these cells did not react to CD19.We have concluded that 2E8 is a B lineage specific marker which can be used in both the diagnosis of B lineage acute lymphoblastic leukemia(ALL )with poor differentiatioii and the differential diagnosis of acute undifferentiated leukemia(AUL).
