人脐血单个核细胞中Nestin阳性细胞表达及其临床意义

摘　　要：目的　观察脐血单个核细胞是否表达nestin抗原及表达量的多少;探讨人脐血单个核细胞(humancordbloodmononuclearcells ,HCMNCs)对神经系统疾病治疗的潜能。方法　采集正常分娩胎儿的脐血。以淋巴细胞密度梯度离心法分离人脐血细胞(humancordbloodcells ,HUCBCs)。用免疫组化法测定HCMNCs及外周血细胞(humanperipheralbloodcells ,HPBCs)中nestin阳性细胞;流式细胞仪(flowcytometer,FCM)分析HCMNCs中nestin阳性细胞的比例及细胞周期。结果　脐血中有nestin阳性细胞,约占HCMNCs的(4 3 2±0 66) %。HCMNCs的G1 0期细胞显著少于外周血而S期细胞则显著多于外周血,G2 M期细胞无显著差异。结论　人脐血单个核细胞确能表达nestin抗原。这将为HCMNCs在神经系统应用提供一定的理论依据。Objective To investigate the expression of nestin in human cord mononuclear cells (HCMNCs) and discuss the relevance of this finding upon future therapeutic application of HCMNCs in neurological diseases. Methods The cord blood from fetus after normal labor was collected and the human umbilical cord blood was isolated by density gradient contrifugation. The nestin-positive cells in the HCMNCs and the peripheral blood were detected by immunocytochemistry. The nestin-positive cell ratio of HCMNCs and cell cycle were analyzed with flow cytometer. Results The nestin-positive cell ratio in HCMNCs was about (4.32±0.66)%. The ratio of G1/0 and S of HCMNCs had a significant difference with that of human peripheral blood cells. Conclusion HCMNCs expressed nestin, which supports the idea that cord blood cells could adopt a neural fate, and they can be applied in central nervous system.
