摘  要:目的 分析新鲜冰冻血浆(FFP)于两种温度下保存1年其凝血因子活性的动态变化,为制定FFP的有效保存温度和保存时间提供依据。方法 FFP分别于- 30℃、- 80℃保存,于0、90、180、2 70、36 0d分别采用一期法测定FⅧ∶C、FⅤ∶C、FⅨ∶C及FⅪ∶C ,用免疫比浊法测定vWF和FⅧ∶CAg。结果 FFP在- 30℃保存1年的FⅧ∶C、FⅤ∶C、FⅨ∶C、FⅪ∶C ,vWF和FⅧ∶CAg均符合质量标准,其中FⅨ∶C及FⅪ∶C ,vWF和FⅧ∶CAg与初期相比差异无显著性;FⅧ∶C与FⅤ∶C显著降低,但高于70 % ;上述各凝血因子在- 80℃保存1年其活性与保存初期相比差异无显著性。结论 FFP于- 30℃保存1年,其凝血因子具有较好的临床止血效果,- 80℃有利于不稳定凝血因子的保存。The aim of our study was to investigate the change of coagulation factors activity in FFP stored at two different temperatures during one year.Methods 16 plasma units were randomly selected,they were aliquoted and frozen at -30℃ and -80℃,respectively. One-stage methods were used to detect the coagulation factors on 0,90,180,270 and 360 d,respectively;the concentration of vWF and FⅧ∶CAg were assayed by immunoprecipitation method.Results All the measured coagulation factors showed an acceptable stability during the total storage at two different temperatures.There were no statistically significant loss for FⅪ∶C,FⅨ∶C,vWF and FⅧ∶CAg when FFP stored at -30℃ during one year.However,for FV∶C and FⅧ∶C a significant reduction was found,although the activity were not reduced below 70%. There were no significant changes for all coagulation factors measured when FFP stored at -80℃ during the total storage. Conclusion Our findings showed that FFP under storage temperature of -30℃ for one year is feasible.It can improve the preservation of labile coagulation factors when FFP stored at -80℃.
