摘 要:Pseudozyma antarctica隶属于担子菌纲。P.antarctica的双质粒共转化系统包括:p Sce I-hyg和p Vectour libre,其中p Sce I-hyg含有hsp70启动子和终止子以及潮霉素抗性基因;p Vectourlibre仅含有hsp70启动子和终止子,以便插入要表达的目的基因。设计引物扩增得到绿色荧光蛋白基因后,插入p Vectourlibre,构建质粒GFP.hsp,将该质粒与p Sce I-hyg共转化P.antarctica;实现了该系统在P.antarctica中的绿色荧光表达。Pseudozyma antarctica is Basidiomycetes speciesbelonging to filamentousfungi.The co- transformation system of P. antarctica includes two plasmids:p Sce I-hyg and p Vectour libre.The plasmid p Sce I-hyg consists of hsp70 promoter,hygromicine gene and hsp70 terminator while the plasmid p Vectour libre only contains hsp70 promoter and terminator in order to be inserted by interesting genes.The specific gene primers were designed based on green fluoresent protein ( GFP) sequence,then GFP gene was obtained by PCR,and inserted into p Vectour libre to achieve the recombinantvector GFP.hsp,which was ready for the co-transformation with p Sce I-hyg.Specific fluorescence can be observed from co-transformation system in P.antarctica using a fluorescent microscope.
